Identificación de especies del género Mycosphaerella en Eucalyptus

ABSTRACT:Several species of the genus Mycosphaerella can cause the foliar disease known as MLB (mycosphaerella leaf blotch) in eucalyptus. In the last years, in Galicia, a high incidence of MLB was observed in Eucalyptus globulus plantations. The analysis of foliar lesions has enabled us to detect several Mycosphaerella species as agents of the disease. Although E. globulus is the more frequently used eucalyptus species in Galician plantations, other Eucalyptus species are also being planted, mostly E. nitens specimens, of which there is not evidence of their susceptibility to MLB, and therefore the Mycosphaerella species involved. In the present work MLB symptomatic material of several eucalyptus species from a nursery in Ares (A Coruña) was analyzed. Mycosphaerella species identification was carried out by analyzing their morphological characteristics (lesions morphology and monosporic cultures) and by molecular techniques in which the fungal DNA was extracted directly from the lesions or from the monosporic cultures. Extracted DNA was amplified and sequenced with primers ITS1F and ITS4. Specific primers for three mycosphaerella species were also used. Results showed that the species more frequently found causing MLB in eucalyptus species was M. parva.
